- Analysis of Inbreeding in a Closed Population of Crossbred SheepMacKinnon, Kathryn Michelle (Virginia Tech, 2003-08-20)Genetic diversity and the effect of lamb and dam inbreeding on multiple traits were analyzed in an 11-yr closed population of sheep established in 1983 and remained closed after 1987, with 50% Dorset, 25% Rambouillet, and 25% Finnsheep breeding to determine selection response for spring fertility. The population had been randomly divided in 1987 into a fall-lambing selection line (S) of 125 ewes and 10 rams, fall-lambing environmental control line (E) of 55 ewes and 5 rams, and a spring-lambing genetic control line (G) of 45 ewes and 5 rams. Inbreeding effects were estimated from 2678 lambs and 556 dams present after the creation of the respective lines. The traits assessed were ewe spring-fertility, lambing date, lamb birth, 60 d, and 120 d weight, and lamb survival to 1, 3, and 14 d. Genetic diversity was assessed by estimating change in inbreeding per generation (ΔF) and effective number of breeding animals (Ne), and parameters derived from gene drop simulations and an iterative procedure developed by Boichard et al. (1997); effective number of founders (fe), effective number of ancestors (fa), founder genome equivalents (fg), and two additional measures of genetic diversity (GD₁, GD2). In order to estimate the diversity available in S and G, three sets of animals from the end of the study and one set of animals at line formation were considered in each line: all lambs born (including dead lambs), all matings (including potential offspring, even if a lamb was not born), and all rams and ewes available at the end of the study and at line formation. At the time of line formation, most of the loss in diversity was due to unequal founder representation. The smaller population of G, as compared to S, caused a greater decrease in diversity due to bottlenecks at line formation. Very little diversity was lost due to additional drift by the time of line formation because selection had not occurred and a random mixing of founders was the goal. Allelic diversity decreased moderately; of the 322 founder alleles, there were 71% in S and 58% in G of rams and ewes (RE) that appeared in at least 50 runs of gene drop. By the end of the study in 1998, the amount of allelic diversity had decreased substantially. Of the alleles possible in RE at the end of the study in S and G, only 6 and 8 %, respectively, appeared in greater than 50 simulations of gene drop. The measures of fe, fa, and fg revealed there was not much additional loss in diversity from the line founders to the end of the study due to unequal founder representation, but there was a larger amount of loss due to bottlenecks and additional drift. The diversity loss was similar, which was the goal of the selection study, when values for RE were compared in S and G. The effects of lamb and dam inbreeding were estimated from REML analysis. Effects of lamb or dam inbreeding were negative but not significant for lambing date or survival to 1, 3, or 14 d. Spring fertility was estimated to decrease by 0.70 ± 0.30 %/% inbreeding of the ewe (P < 0.01), which seems even greater since the average spring fertility was only 47.5 %. Effects of lamb inbreeding on birth, 60-d, and 120-d weights were -0.012 ± 0.006 (P < 0.05), -0.045 ± 0.020 (P < 0.05), and -0.130 ± 0.034 kg/% (P < 0.01), respectively. Dam inbreeding had smaller effects on birth, 60-d, and 120-d weights of -0.008 ± 0.010 (ns), -.033 ± 0.034 (ns), and -0.087 ± 0.056 (P < 0.1) kg/%, respectively.
- Analysis of newborn calf body measurements and relationship of calf shape to sire breeding values for birth weight and calving easeNugent, Russell Arthur (Virginia Tech, 1990)These studies were conducted to define calf body shape, to test for relationships between calf shape and sire expected progeny differences for birth weight (BWTEPD) and first-calf calving ease (CEEPD) and to evaluate the efficacy of calf shape as a selection criterion for reduction of calving difficulty. Birth weight (BW), head circumference (HC), hip width (HW), shoulder width, body length (BL), cannon circumference (CC), cannon length (CL) and heart girth (HG) were measured at birth on 1,016 calves sired by Angus, Polled Hereford and Simmental bulls. In the Angus- and Polled Hereford-sired calves, sire effects were present for BW-adjusted residuals of CC. Additionally, HW, BL and HG residuals differed among half-sib groups for the Polled Hereford-sired calves. Thus, calf body measure differences independent of BW were in part attributable to sire. Multivariate factor analysis was used to identify underlying skeletal width and frame dimensions of calf shape in both breeds. After adjustment of body measures for differences in BW, a positive relationship of BWTEPD with HC and CC and a negative relationship between CEEPD and CC existed. Further adjustment for BWTEPD removed effects of CEEPD on CC. Thus, BWTEPD influenced calf shape independent of BW, but shape was not related to CEEPD independent of BWTEPD. Simmental bulls were divergently selected on CEEPD relative to BWTEPD so that body measures of calves from sires whose progeny tended to be born with more or with less dystocia than expected from BWTEPD could be obtained. Differences in CL and CC at constant BW were in part attributable to sire. Underlying shape factors were similar to those of the other breeds. Sire BWTEPD was positively related to CC and HC independent of BW. However, a relationship between body measures and CEEPD existed only at constant BW and BWTEPD. Calf shape independent of BW was also not different among calves born unassisted and assisted. Estimates of heritability, repeatability and birth to weaning relationships for each BW-residual body measure were generally not significant. Overall, sire BWTEPD was related to calf shape independent of BW. However, increases in skeletal dimensions were not related to either observed calving difficulty nor sire CEEPD independent of BW. Selection for reduced calving difficulty should not be based on calf body shape.
- Association between FAMACHA scores and fecal egg counts in Katahdin lambsNotter, David R.; Burke, Joan M.; Miller, James E.; Morgan, James L. M. (2017-03)The FAMACHA system was introduced to the U.S. just over 10 yr ago to allow selective deworming of lambs with anemia associated with Haemonchus contortus and retard the development of anthelmintic resistance. The FAMACHA system was initially developed as a predictor of packed cell volume (PCV), but correlations between FAMACHA and fecal egg counts (FEC) have also been reported. It is important to understand factors that influence FAMACHA scores among farms to improve management of gastrointestinal nematodes. The objectives of this study were therefore to quantify associations between FAMACHA scores, FEC, BW, and age in Katahdin lambs at 2 different measurement times in 8 flocks in the eastern U.S., and to assess consistency of relationships between FAMACHA and FEC among flocks. Data came from 1,644 Katahdin lambs from 7 flocks sampled at approximately 90 d of age, and 1,295 lambs from 6 flocks sampled at approximately 120 d of age over a 5 yr period. Residual correlations among log-transformed FEC (LFEC), FAMACHA scores, BW, and lamb ages at each measurement time were determined. Repeatability of each variable was also determined as residual correlations among repeated measures. At both 90 and 120 d of age, correlations of FAMACHA scores with LFEC and BW were significant (P < 0.001), but numerically modest (0.25 and -0.16, respectively at 90 d; 0.31 and -0.16, respectively at 120 d), demonstrating that higher FAMACHA scores were associated with higher FEC and more likely to be observed in lighter lambs. A small negative correlation was observed between FAMACHA score and lamb age (r = -0.05, P = 0.05, 90 d; r = -0.11, P < 0.001, 120 d) indicating that younger lambs were more likely to have elevated FAMACHA scores. Thus, younger and lighter lambs will likely be more susceptible to parasitism and may need to be managed more diligently than older or heavier lambs. In addition, FAMACHA scores have potential to improve breeding value estimates in programs designed to genetically improve parasite resistance.

Ewes were born in 1979 or 1980; no R ewes were born in 1980. Ewes were bred in April, 1980 (APR80); November, 1980 (NOV80); August, 1981 (AUG81); May, 1982 (MAY82); and April, 1983 (APR83). Ewes born in 1980 entered the study in NOV80. - Budget Analysis of Spring, Fall with Winter Clean-up, and High-Fertility Fall Lambing Systems in a Simulated Fixed Forage ResourceAndrew, Shelley Lewis Jr. (Virginia Tech, 1998-08-03)A successful business needs to generate enough cash to cover expenses, current debt, and family living expenses, pay interest on owned and borrowed capital, maintain productivity, and earn a reasonable return for the operator. Income from sheep production is generally only part of a total farm and nonfarm income. Thus options, opportunity costs, and decisions regarding the sheep production enterprise are not isolated; they affect other agricultural enterprises. Sheep production consistently returns profits to producers, which makes it an enticing agricultural enterprise. There are advantages in raising sheep in Virginia, such as abundant, high-quality forage, moderate climate, pasture improvement, and good access to markets with high demand for lamb. The disadvantages to sheep production are unavailable and inexperienced labor and operators, predators, and inconsistent market demand and supply. Sheep producers have the opportunity to choose which lambing system fits their existing operations and lifestyle. The use of economic analysis enables operator to make sound business management decisions. To compare different lambing systems (spring, fall with winter clean-up, and high-fertility fall) in a systematic way, a simulation model was constructed with a fixed forage resource of 50 acres of pasture including typical Virginia mountain pasture plus various amounts of fescue for stockpiling. The simulation included a production calendar; nutritional requirements for ewes, lambs, and artificially reared triplets; growth rates for lambs; lambing distributions; forage growth; and enterprise budgets including income, costs, and returns. A economic analysis was performed for each lambing system with average prices or with plus or minus one standard deviation for prices of corn, SBOM, and market lambs, and price differentials for market lambs across lambing seasons. Comparisons of each lambing systems produced various results. In spring lambing, only 78 ewes could be maintained on the fixed forage resource, while the fall with winter clean-up and high-fertility fall lambing system each had 115 ewes. This result occurred because of limited forage in July and August and higher nutrient requirements for spring lambing in those months. The overall nutrient requirements were higher in the fall with winter clean-up and high-fertility fall lambing than in spring lambing as a result of the increased ewe and lamb numbers. Concentrate consumption by lambs was also greater for fall with winter clean-up and high-fertility fall lambing than for spring lambing because of the increased numbers of lambs. Because of the low number of ewes and lambs, spring system produced the most hay. Labor costs were highest in fall with winter clean-up lambing because of the two lambing seasons. In the economic analysis system, each lambing was compared. With 10-year average prices for market lambs, corn, and SBOM, high-fertility fall lambing had the greatest income ($17,467), followed by fall with winter clean-up lambing ($14,695), and spring lambing ($10,358). This result occurred because high-fertility fall and fall with winter clean-up lambing had more lambs sold at higher market lambs prices than spring lambing. With 10-year average prices for market lambs, corn, and SBOM, high-fertility fall lambing had the highest cost ($7,935), followed by fall with winter clean-up lambing ($7,360), and spring lambing ($6,084). This was the result of increased ewe and lamb numbers in high-fertility fall and fall with winter clean-up lambing than spring lambing. High-fertility fall lambing had the greatest returns ($6,210), followed by fall with winter clean-up lambing ($4,025), and spring lambing ($2,028). On a fixed forage resource, increasing fertility in fall lambing clearly results in increased returns. In this model, forage availability controlled the number of ewes that a lambing system can have because of limited summer growth and had a major impact on profits. Conclusions of Tolman (1993) differed from those found within this thesis. On a per ewe basis, she found that spring lambing to yielded the highest returns whereas this thesis found that high-fertility fall lambing yielded the highest returns. A key difference between this study and that of Tolman (1993) was after weaning this thesis feed fall lambs stockpiled fescue and she feed fall lambs feed in dry lot.
- Characterization of seasonal reproduction in Virginia Tech Selection Line, St. Croix, and Suffolk ewesJordan, Katherine Mead (Virginia Tech, 2008-08-04)This dissertation research contained three studies. The first two studies were conducted to investigate the ability of ewes to rebreed while lactating during seasonal anestrus. Breeds studied included the Virginia Tech Out-of-season (OOS) Line, which is a wool line genetically selected to lamb in the fall, and the St. Croix, a hair breed of tropical origin thought to be lowly seasonal. When January-lambing ewes were exposed to rams while lactating in April, significantly more OOS than St. Croix ewes were marked by rams in the first 21 d and total 39 d of ram exposure (58.3 vs. 8.7%, P = 0.0003 and 95.8 vs. 43.5%, P < 0.0001). Percentages of ewes diagnosed pregnant (53.2%) and percentages of ewes lambing (41.3%) were not different between breeds. When March-lambing OOS ewes were exposed to rams while lactating in May, 52.9% of ewes were marked though only 20% of ewes exposed to rams gave birth to viable lambs. Both OOS and St. Croix ewes appear to be well suited to accelerated production systems involving 7 to 8 mo lambing intervals. However, reduction of lambing intervals to 6 to 7 mo appeared to have detrimental effects on fetal survival in OOS ewes. In a third study, alterations in endocrine profiles associated with differing degrees of hypothalamic sensitivity to estradiol-negative feedback and changing daylength in OOS, St. Croix, and Suffolk ewes in the absence of rams were investigated for 1 yr. The results show for the first time that based on progesterone profiles from intact ewes, St. Croix ewes do not have shorter anestrous periods than ewes of wool breeds, as previously thought. Based on luteinizing hormone profiles from ovariectomized ewes treated with estradiol implants, the duration of luteinizing hormone inhibition was shorter in OOS than Suffolk ewes (68 vs. 170.2 d, P = 0.02), but was not different from that found in St. Croix ewes (124.8 d). Specific roles for thyroxine and prolactin in timing the breeding season could not be assigned. This study was the first known use of the ovariectomized, estradiol-implanted ewe model to compare degree of reproductive seasonality in different breeds.
- Characterizing physiological and genetic differences in the early immune response to Haemonchus contortus in resistant and susceptible sheepBowdridge, Scott Alexander (Virginia Tech, 2009-10-12)This dissertation compares immune responses of resistant and susceptible sheep to infection with Haemonchus contortus during the peri-parturient period and larval stage of infection. Identification of immunological events resulting in parasite resistance in St. Croix hair sheep may provide better targets for differential gene expression analysis and eventual discovery of selectable markers for parasite resistance. Antibody levels of hair ewes and composite Dorset x Finnsheep-Rambouillet wool ewes were measured during breeding and again after parturition. Results demonstrated that hair ewes had higher levels immunoglobulin-A after infection and maintained a higher level of circulating antigen-specific antibody when compared to wool ewes. To characterize immune responses to the larval stage of infection, hair and wool lambs were sacrificed at 0, 3, 5, and 7 d after infection with H. contortus. Neutrophil migration to abomasal mucosa and lymph node development were higher in hair sheep than in wool sheep. Gene expression analysis indicated no difference in the abomasal lymph node as both breeds expressed a general T-helper cell type 2 (TH2) response. However, profound differences in TH2 responses were observed in the abomasal mucosa, where hair sheep expressed more IL-4, -13 and -33 than wool sheep. These data thus document the presence of immunological differences between the breeds. Immune responses to larval parasite infection in wool sheep are generally suppressed and may increase the magnitude and duration of infection whereas immune responses to larval infection in hair sheep was more robust and more strongly polarized towards a TH2
- Combining ability, protein, heterosis, and prediction of F₁ performance with RFLPs in a diallel of maizeBall, Dale Warren (Virginia Tech, 1994-07-06)Improving protein quality and identifying superior inbreds and hybrids are significant challenges in commercial maize breeding programs. These two problems were addressed in separate studies on inbreds and hybrids from a complete diallel cross of 12 elite proprietary inbred lines of maize evaluated in field trials in two locations for two years. One of the inbreds (WI) was a novel source of high quality protein obtained from Wilson Seeds, Inc. in Harlan, Iowa. In the first study, diallel analyses were used to study combining ability and types of gene action important in the inheritance of protein content, grain yield, grain moisture at harvest, time to silk, kernel hardness, and density. General combining ability (GCA) and specific combining ability (SCA) effects were highly significant for all traits indicating presence of both additive and non-additive effects, respectively. Reciprocal effects (REe), often assumed to be absent in maize diallel studies, were significant for grain yield and protein concentration, suggesting that choice of female parent may be important for these traits. Ratios expressing the relative importance of GCA and SCA indicated that protein concentration is controlled primarily by additive gene action. In the second study, restriction fragment length polymorphism (RFLP) data were obtained for the 12 inbreds using 42 genomic clones each with four restriction enzymes. Modified Roger's distances were calculated and used in cluster analyses for heterotic grouping of the inbreds. Two measures of level of heterozygosity and hybrid value were evaluated as means of predicting Fl performance of hybrids in the complete diallel set of hybrids and in groups of hybrids representing crosses between and within heterotic groups. Results from this study confirm those of previous investigations with respect to prediction of hybrid performance when comparable groupings of crosses between related and unrelated lines were evaluated. This study further indicates that RFLPs may also be useful for prediction of hybrid performance in situations typical of early generations of many maize breeding programs where recombinant inbreds are testcrossed to a common tester inbred.
- Comparison of expected and actual progeny growth differences in crossbred calvesMahrt, Garry Sonn (Virginia Tech, 1987-06-05)The growth performance of Angus-Polled Hereford F1 calves was compared to the expected progeny differences (EPD) estimated by the American Polled Hereford Association to evaluate the usefulness of EPD estimates for improving commercial beef production. Polled Hereford sires were selected from the high accuracy sires listed in the American Polled Hereford Association's sire summary. Four types of sires were used: 1) Sires with high yearling weight and high maternal EPD's; 2) Sires with high yearling weight and low maternal EPDâ s; 3) Sires with low yearling weight and high maternal EPD's; and 4) Sires with low yearling weight and low maternal EPD's. The resulting progeny were weighed at birth, approximately 135 d, weaning and approximately 1 yr. Hip heights were also measured at weaning. Linear regressions of progeny performance on EPD indicated that birth and yearling weights were accurately predicted by their respective EPD estimates. Weight and height at weaning were not accurately predicted by weaning weight EPD. Partial regressions of progeny performance on yearling weight and maternal EPD's were also calculated. Regression of progeny performance on yearling weight EPD resulted in .16 ±.09 kg/kg for 135-d weight, .28 ± .09 kg/kg for weaning weight, .073 ± .020 cm/kg for weaning hip height and .93 ±.19 kg/kg for yearling weight. Comparable regression values for maternal EPD were .44 ± .18 kg/kg, .31 ± .17 kg/kg, .100 ± .040 cm/kg and .22 ± .36 kg/kg for 135-d, weaning weight, weaning hip height and yearling weight, respectively. These results indicate that maternal EPD values predicted some differences in preweaning growth that were not predicted by yearling weight EPD. The progeny performance data was also used to calculate independent EPD estimates for birthweight, weaning weight, and yearling weight. Correlations between these EPD estimates and those made by the American Polled Hereford Association were .49 for birthweight, .26 for weaning weight and .66 for yearling weight. The low correlation between weaning weight EPD estimates was significantly below expectation and indicates that the American Polled Hereford Association estimates were inadequate for predicting preweaning growth differences in these crossbred calves.

- Correlation of predicted breeding values across environments in the presence of selection for direct and maternal breeding valuesDiaz-Martin, Clara (Virginia Tech, 1992-08-05)A simulation approach was used to determine the effects of multitrait selection on the correlations of sire direct and maternal predicted breeding values across environments. True and predicted direct and maternal breeding values (BV) of sires were simulated for sires evaluated independently in two different environments. Prediction error variances and covariances among direct and maternal BV within environments were required for the simulation. To obtain the necessary input parameters, a variety of MME coefficient matrices were created and inverted to inspect relationship among accuracies and correlations of prediction errors in sire evaluation models. An empirical prediction equation to predict the necessary prediction error covariances was obtained. Divergent, directional and random multitrait selection was then practiced using direct and maternal predicted BV as selection criteria. Samples of 40 sires were randomly obtained from each selected population. Observed correlations between direct and maternal predicted BV across environments were compared to expectations derived from univariate distribution theory. Selection definitely affected the expectations. However, the adjustment developed from univariate theory appeared to accommodate the effect of selection in these expectations.
- Designing and modeling high-throughput phenotyping data in quantitative geneticsYu, Haipeng (Virginia Tech, 2020-04-09)Quantitative genetics aims to bridge the genome to phenome gap. The advent of high-throughput genotyping technologies has accelerated the progress of genome to phenome mapping, but a challenge remains in phenotyping. Various high-throughput phenotyping (HTP) platforms have been developed recently to obtain economically important phenotypes in an automated fashion with less human labor and reduced costs. However, the effective way of designing HTP has not been investigated thoroughly. In addition, high-dimensional HTP data bring up a big challenge for statistical analysis by increasing computational demands. A new strategy for modeling high-dimensional HTP data and elucidating the interrelationships among these phenotypes are needed. Previous studies used pedigree-based connectetdness statistics to study the design of phenotyping. The availability of genetic markers provides a new opportunity to evaluate connectedness based on genomic data, which can serve as a means to design HTP. This dissertation first discusses the utility of connectedness spanning in three studies. In the first study, I introduced genomic connectedness and compared it with traditional pedigree-based connectedness. The relationship between genomic connectedness and prediction accuracy based on cross-validation was investigated in the second study. The third study introduced a user-friendly connectedness R package, which provides a suite of functions to evaluate the extent of connectedness. In the last study, I proposed a new statistical approach to model high-dimensional HTP data by leveraging the combination of confirmatory factor analysis and Bayesian network. Collectively, the results from the first three studies suggested the potential usefulness of applying genomic connectedness to design HTP. The statistical approach I introduced in the last study provides a new avenue to model high-dimensional HTP data holistically to further help us understand the interrelationships among phenotypes derived from HTP.
- Developing Breeding Objectives for Targhee SheepBorg, Randy Charles (Virginia Tech, 2004-06-08)Breeding objectives were developed for Targhee sheep at different levels of prolificacy and triplet survival. Economic weights (EW) were derived for estimated breeding values (BV) from National Sheep Improvement Program genetic evaluations for 120 d weaning weight (WW), maternal milk (MM), yearling weight (YW), fleece weight (FW), fiber diameter (FD), staple length (SL), and prolificacy (PLC; lambs born/100 ewes lambing). A commercial flock was simulated, accounting for nonlinear relationships between performance and profit. Ewes were assumed mated to sires of specified BV and profit was derived from lifetime performance of lambs and replacement females from that lamb crop. Economic weights were determined as change in profit from use of sires with BV that were one additive standard deviation above the mean for each trait [1.98 kg for WW, 1.62 kg for MM, 2.90 kg for YW, 0..36 kg for FW, 0.99 microns for FD, 0.74 cm for SL, and 17.58 lambs/100 ewes for LC], while holding all other BV at breed average. Separate breeding objectives were derived for different ways of meeting increased nutrient needs (P = purchase hay, R = rent pasture, and L= limited flock size) and for different market lamb values (D = discounting lamb value for heavy weights, ND = no discount for heavy lambs). Based on replicated simulations, relative EW did not vary with prolificacy or triplet survival (P > 0.15) but were affected by feed costs and lamb market values (P < 0.01). Selection indexes were derived within and across simulated scenarios, and correlation (r) among indexes of > 0.90 indicated that an index could be used across multiple scenarios with little loss of selection efficiency. Indexes derived within feed cost scenarios (P, R, and L) and lamb value scenarios (D, ND) were strongly intercorrelated (r > 0.97). Correlations among average indexes for feed cost scenarios (0.97 for R and P, 0.70 for R and L; 0.85 for P and L) indicated that two feed cost scenarios could be used depending on whether winter forage was limited (L) or not (NL). The correlation between average indexes for these two scenarios was 0.78. Indexes were presented for combinations of feed cost and lamb value scenarios. Two indexes were suggested, representing the scenarios that apply to a large portion of Targhee producers. These indexes were for discounting heavy lambs with limited winter forage (D-L: 1.0 WW + 0.14 MM __ 0.76 YW + 1.22 FW __ 0.36 FD - 0.09 SL + 0.25 LC) and discounting heavy lambs with additional available forage (D-NL: 1.0 WW + 0.24 MM __ 0.34 YW + 1.65 FW __ 0.41 FD - 0.14 SL + 0.33 LC). For a standardized selection differential of one for the index, the expected changes in mean index value were $2.17 and $1.92 per ewe per generation for D-L and D-NL, respectively.
- Differential gene expression and immune regulatory mechanisms in parasite-resistant hair and susceptible wool sheep infected with the parasitic nematode, Haemonchus contortusMacKinnon, Kathryn Michelle (Virginia Tech, 2007-07-23)Among sheep producers, the parasitic nematode Haemonchus contortus is a major animal health concern. Caribbean hair sheep are more resistant than conventional wool breeds to this blood-feeding, abomasal parasite. Our objective was to determine differences in the immune response associated with parasite-resistant hair and susceptible wool lambs infected with 10,000 H. contortus and in uninfected controls. Animals were sacrificed and abomasum and lymph node tissues were collected at 3 or 27 days post-infection (PI), and for controls on day 17, 27, or 38 relative to d 0 of infected animals. Blood and fecal samples were collected throughout the study. Lower fecal egg counts, higher packed cell volumes, and heavier lymph nodes of infected hair compared to wool lambs, suggests hair lambs have increased parasite resistance. Greater tissue infiltration of eosinophils (P < 0.05) was observed in hair compared to wool sheep by 3 days PI, with no breed differences in globule leukocytes. Total serum IgA and IgE were greater in control hair versus wool sheep (P < 0.05). After 3, 5, and 21 of infection, total serum IgA (P< 0.05), total lymph node IgE (P < 0.01), but not total serum IgE were greater in hair sheep compared to wool sheep. Gene expression was measured between hair and wool lambs for abomasal and lymph node tissues using bovine cDNA microarrays and real-time RT-PCR. Microarray analysis revealed cell survival, endosome function, gut motility, and anti-coagulation pathways are important in abomasal and lymph node tissues during H. contortus infection. Immune genes, including IL-4, IL-4 Ra, IL-12 Rb1, and IL-12 Rb2, are also highly represented in abomasal or lymph node tissue of infected animals. Eleven genes were evaluated using real-time RT-PCR and included TH1 and TH2 cytokines, cytokine receptors, and IgE. Parasite infection leads to increased expression of IL-13 and IgE in both tissues and breeds when compared to control animals. Breed comparison of gene expression shows resistant hair sheep produce a stronger modified TH2-type immune response during infection. Differential cell infiltration, antibody production, and regulation of TH2 cytokines between breeds may be partially responsible for differences in parasite resistance.
- Direct and correlated responses to selection for autumn lambing in sheepAsadi-Fozi, Masood; Bradford, Heather L.; Notter, David R. (2020-10-02)Background Seasonal reproduction limits productivity, flexibility, and profitability in commercial sheep production. Hormonal and (or) photoperiodic manipulation can be used to control estrous cycles in sheep and reduce limitations that are imposed by the seasonal anestrous but are often impractical or incompatible with the extensive management systems preferred for ruminant livestock. Thus, the current study investigated the use of selection to improve realized fertility (i.e., the proportion of ewes that lambed) following an out-of-season spring joining period (May and June) in a crossbred sheep population. Results Over 17 years, estimated breeding values (EBV) for fertility in selected (S) ewes increased by 0.175 (0.01 per year). The mean EBV for fertility of S ewes was greater than that of control ewes by year 10 (P = 0.02), and the fertility of adult (≥ 3 years old) ewes reached 0.88 ± 0.05 by year 17. Lambing began approximately 140 days after the introduction of rams, and 64% of the S ewes that lambed did so in the first 17 days of the potential lambing season, which indicated that most of the S ewes were cycling at the time of ram introduction and were not induced to cycle by the introduction of breeding males (i.e., the so-called “ram effect”). Animals in the S line had modest increases in body weight and scrotal circumference. A modest negative trend in the additive maternal effect on birth weight was observed but was reversed by additional selection on EBV for maternal birth weight. The heritability of litter size in autumn lambing was low (0.04) and could potentially limit the response to selection for this trait. Conclusions Selection improved realized ewe fertility in out-of-season mating, with absolute increases of approximately 1% per year in the percentage of joined ewes that lambed in the autumn. Genetic antagonisms with other performance traits were generally small. A modest antagonism with maternal breeding values for birth weight was observed but it could be accommodated by selection on EBV for maternal birth weight. Our results support results of previous studies that indicate that these selected ewes had one of the shortest seasonal anestrous periods reported for temperate sheep breeds and that spring-lambing lactating ewes from the selection line were capable of relatively rapid rebreeding in the spring.
- The effect of dietary vitamin E on the humoral and cell-mediated immune response of pigs housed at different environmental temperatures or weaned at various agesBonnette, Edward Dwain (Virginia Polytechnic Institute and State University, 1988)A set of experiments were conducted to evaluate the antibody response, serum vitamin E level, cortisol concentration and performance of pigs weaned at three ages (21, 28 or 35 d) and fed diet containing either 11 or 220 IU/kg diet recommended level of vitamin E. Supplemental dietary vitamin E (220 IU) increased the concentration of serum vitamin E but, did not affect performance, cortisol concentrations or the antibody response. As weaning age increased, weekly performance increased linearly as did cortisol levels. Animals weaned at 35 d age had the largest primary antibody response, but this difference was not observed for the secondary response. A second set of experiments evaluated effects of four dietary vitamin E levels (11, 110, 220 and 550 IU/kg feed) on the humoral and cell-mediated immune response and performance of 4 wk old weanling pigs housed at one of two nursery temperatures (19 or 30°C). Performance was greater for pigs housed at 19°C compared with pigs housed at 30°C, but mitogen stimulation indices of white blood cells, plasma cortisol levels, and antibody titers were similar. Serum and liver vitamin E levels linearly increased with increasing dietary vitamin E level, but performance, cortisol, antibody levels and mitogen induced stimulation indexes were not affected by supplemental vitamin E levels. In the third experiment, sows fed a NRC level of vitamin E demonstrated little fluctuation in serum vitamin E concentration during a 5 wk lactation period. There was a high concentration of vitamin E in colostrum, followed by a sharp decrease in milk vitamin E concentration after the first week of lactation and remained constant for the next four weeks. Piglet serum was initially low in vitamin Eat 1 d of age but increased with time peaking about week 3. These experiments suggest that supplementing dietary vitamin E above the levels recommended by the NRC to baby pigs (which nursed sows fed NRC recommended levels of vitamin E) will not influence cell-mediated or humeral immune response, performance parameters or cortisol levels when pigs were weaned at various ages or exposed to environmental temperature changes.
- The Effect of Maternal and Fetal Inbreeding on Dystocia, Calf Survival, Days to First Service and Non-Return Performance in U.S. Dairy CattleAdamec, Vaclav (Virginia Tech, 2002-01-07)Intensive selection for increased milk production over many generations has led to growing genetic similarity and increased relationships in dairy population. In the current study, inbreeding depression was estimated for number of days to first service, summit milk, conception by 70 days non-return, and calving rate with a linear mixed model (LMM) approach and for calving difficulty, calf mortality with a Bayesian threshold model (BTM) for categorical traits. Effectiveness of classical and unknown parentage group procedures to estimate inbreeding coefficients was evaluated depending on completeness of a 5-generation pedigree. A novel method derived from the classical formula to estimate inbreeding was utilized to evaluate completeness of pedigrees. Two different estimates of maternal inbreeding were fitted in separate models as a linear covariate in combined LMM analyses (Holstein registered and grade cows and Jersey cows) or separate analyses (registered Holstein cows) by parity (1-4) with fetal inbreeding. Impact of inbreeding type, model, data structure, and treatment of herd-year-season (HYS) on magnitude and size of inbreeding depression were assessed. Grade Holstein datasets were sampled and analyzed by percentage of pedigree present (0-30%, 30-70% and 70-100%). BTM analyses (sire-mgs) were performed using Gibbs sampling for parities 1, 2 and 3 fitting maternal inbreeding only. In LMM analyses of grade data, the least pedigree and diagonal A matrix performed the worst. Significant inbreeding effects were obtained in most traits in cows of parity 1. Fetal inbreeding depression was mostly lower than that from maternal inbreeding. Inbreeding depression in binary traits was the most difficult to evaluate. Analyses with non-additive effects included in LMM, for data by inbreeding level and by age group should be preferred to estimate inbreeding depression. In BTM inbreeding effects were strongly related to dam parity and calf sex. Largest effects were obtained from parity 1 cows giving birth to male calves (0.417% and 0.252% for dystocia and calf mortality) and then births to female calves (0.300% and 0.203% for dystocia and calf mortality). Female calves from mature cows were the least affected (0.131% and 0.005% for dystocia and calf mortality). Data structure was found to be a very important factor to attainment of convergence in distribution.
- Effects of birth-rearing type on weaning weights in meat sheep are systematically associated with differences in mean performance among flocksNotter, David R.; Brown, Daniel J. (2015-07-03)Background Adjustment of body weights for systematic environmental effects such as dam age and litter size is essential for accurate prediction of breeding values in meat sheep and often accomplished by pre-adjusting records using simple multiplicative adjustment factors, which are derived as ratios of least-squares means of weights of lambs in target and reference classes. However, increasing use of multibreed genetic evaluations that incorporate data from both purebred and commercial flocks has generated concerns regarding the ability of simple additive or multiplicative adjustment factors to properly correct for environmental effects in flocks that differ widely in mean performance. Thus, consistency of adjustment factors across flocks and systematic effects of the level of flock performance on these factors were evaluated using data from the US National Sheep Improvement Program. Methods We used birth and weaning weights of lambs from 29 flocks that had at least 500 records per flock and represented several terminal-sire sheep breeds. Effects of lamb sex, dam age class and litter size on birth weights, and of dam age class and combined effects of type of birth and rearing on weaning weights were evaluated. Interactions between these effects and flock were assessed. Bias associated with different adjustment protocols was evaluated for high- and low-performance flocks. Results Effects of litter size and differences between yearling and adult dams varied (P < 0.001) among flocks. For weaning weights, additive adjustment factors were not associated with the level of flock performance, but multiplicative adjustment factors were strongly and inversely related to flock means for weaning weights (W). Flock-specific adjustment factors (F = αWβ) reduced bias in adjusted weaning weights associated with differences in flock performance. By contrast, simple multiplicative adjustment factors were appropriate to adjust birth weights. Conclusions Differences in weaning weights among single, twin, and triplet lambs were inversely related to the level of flock performance. Use of simple multiplicative adjustment factors led to adjustment bias when applied across flocks with large differences in mean performance. This bias was reduced by using additive adjustment factors or multiplicative factors that were derived as simple exponential functions of flock means for weaning weight.
