- Acetone, Butanol, and Ethanol (ABE) Production from Food Waste via Clostridium beijerinckiiPoe, Nicholas Edward (Virginia Tech, 2020-01-28)Annually, approximately 150 million metric tonnes of food goes to waste in the U.S., potentially causing economic loss and environmental pollution. Fermentation of food waste to produce acetone, butanol and ethanol (ABE) via fermentation has the potential to valorize food waste by producing value-added chemicals. However, the composition of food wastes from different sources vary, which affects ABE fermentation performance and hinders the commercialization of food waste fermentation. The objective of this study is to investigate the compositional variation of food waste collected weekly for 16 weeks (a total of sixteen samples) and determine how this variation affects ABE fermentation performance. Samples collected from Southgate Center, a food processing facility operated by Virginia Tech Dining Services, was characterized for use as a feedstock for ABE fermentation. Water, sugar, starch, fiber, protein, fat and ash concentrations in each of food waste samples were determined. ABE fermentation of these wastes was performed using Clostridium beijerinckii via batch fermentations. Correlations of ABE and butanol yields with the individual components of food waste composition were performed to better understand which components are key to ABE fermentation. Overall, this study demonstrated the feasibility of using food waste as a viable feedstock for ABE fermentation and investigated the effect of variation of food waste composition on the ABE fermentation performance. In the 16 collected samples, each major compositional attribute exhibited high variability. The concentration of total soluble sugar, defined as glucose, fructose, sucrose for the purposes of this experiment, ranged from 0.5 to 53.5% (dry basis) among different food waste samples. The concentration ranges of total starch, neutral detergent fiber (NDF), crude protein, crude fat and ash were 0 to 23.4% (dry basis), 0.6 to 25.8%, 5.5 to 21.2%, 0.1 to 37.9%, 1.4 to 13.7%, respectively. The high variation of food waste composition resulted in a high variation of ABE yield when these food wastes were subjected to fermentation by C. beijerinckii. The total ABE concentration following fermentation ranged between 6.9 to 17.0 g/L with an average value of 13.2 g/L. ABE and butanol concentrations are positively correlated with starch and equivalent glucose, i.e., the sum of initial free glucose and glucose that could be theoretically hydrolyzed from starch and sucrose during fermentation, but is negatively correlated with NDF concentrations.
- Alternative Waste Treatment System for Poultry Processing PlantsRoshdieh, Rana (Virginia Tech, 2010-08-04)The objective of this research was to design an alternative wastewater treatment system for turkey processing plants to recover energy and reduce N and P to allowable discharge levels. The objective included: 1. Determine the quantity and quality of biogas produced from the turkey processing wastewater (TPW) and COD reduction efficiency. 2. Design a waste treatment system and validate proof of concept for simultaneous P and N removal with a goal of attaining effluent concentrations of 0.1 mg/L and 4 mg/L, for P and N, respectively. A lab-scale complete mixed anaerobic digester was used for turkey processing wastewater (TPW) digestion and biogas recovery running for 6 months. Along with the anaerobic digester, a two-sludge system called A2N-SBR consisting of an anaerobic-anoxic sequencing batch reactor and an attached growth post-nitrification reactor was added for biological nitrogen and phosphorus removal running for 3 months. Biogas production yields of 778 + 89 mL/gVSadded and 951.30 mL/g COD were obtained through anaerobic digestion. Also, an energy balance was conducted on a pilot scale digester for a turkey processing plant with wastewater production of 2160 m3/d and using a combined heat and power (CHP) enginefor conversion of biogas to heat and electricity. Although the biogas yield achieved in a complete mixed reactor was relatively lower than yields obtained in previous studies using reactors such as UASB, still a complete mixed reactor can be a good choice for biogas recovery from TPW and can be used for codigestion with some specific turkey processing byproducts for biogas recovery. Nitrogen and phosphorus removal in the A2N-SBR system were 47% and 75%, respectively, and during the study the nitrogen and phosphorus removal mean concentration in effluent did not meet the nutrient limits specified in the objectives. Average TP and TN in the effluent were 3.2 mg/L and 137 mg/L, respectively. Throughout the study, the nitrification reactor biofilm was not completely developed. Incomplete nitrification and poor settling might be the reasons that quality obtained in effluent was low. To improve the process condition in A2N-SBR, online monitoring of pH, dissolved oxygen (DO) and oxidation reduction potential (ORP) can help to optimize each stage in the SBR and stages duration can be set based on the results.
- Ammonia Emissions from Dairy Manure Storage Tanks Affected by Diets and Manure Removal PracticesLi, Lifeng (Virginia Tech, 2009-08-07)The objectives of this study were to determine: 1) ammonia emission rates from stored scraped and flushed manure from dairy cows fed either normal or low N diet; and 2) seasonal effects on ammonia emission rates from stored scraped and flushed dairy manure. Four pilot-scale tanks were used for manure storage with different treatments - scraped manure for normal diet (NS), flushed manure for normal diet (NF), scraped manure for low N diet (LS), and flushed manure for low N diet (LF). The first part of the study lasted for 1 month and four treatments were all investigated; the second part of the study lasted for 12 months and two tanks with treatments NS and NF were investigated. Dynamic flux chambers and a photoacoustic gas analyzer were used to measure ammonia emission rates. There was no significant change of the N content of manure as the dietary N content is reduced (from 17.8% to 15.9% crude protein). However, ammonia emission rates from manure storage tanks were reduced by 33% (from 27.4 ± 38.1 to 18.4 ± 21.9 mg m⁻²h⁻¹; P<0.0001 based on paired t-test). Flushing manure reduced emission rates by 72% compared to scraping manure (from 35.6 ± 39.6 to 10.1 ± 8.2 mg m⁻²h⁻¹; P<0.0001 based on paired t-test). Ammonia emission rates for NS, NF, LS and LF were 43.9 ± 48.0, 10.9 ± 8.7, 27.4 ± 27.3, and 9.3 ± 7.8 mg m-2 h-1, respectively. The chamber headspace temperature for NS, NF, LS and LF were 26.0 ± 6.9, 25.8 ± 6.8, 26.6 ± 6.5, and 27.2 ± 6.7 °C, respectively. The manure pH for NS, NF, LS, and LF were 6.3 ± 0.1, 6.4 ± 0.3, 6.4 ± 0.1, and 6.1 ± 0.1, respectively. Both dietary N reduction and manure flushing are recommended to reduce ammonia emission rates from dairy manure storage tanks. Ammonia emission rates were higher in summer and fall, due to higher air temperature and higher manure pH. The pH of scraped manure was 7.2 ± 0.6, 6.7 ± 0.2, 6.5 ± 0.3 and 7.0 ± 0.3 for fall, winter, spring and summer, respectively. The pH of flushed manure was 6.8 ± 0.4, 6.7 ± 0.4, 6.4 ± 0.3 and 6.8 ± 0.4 for fall, winter, spring and summer, respectively. Ammonia emission rates from scraped manure for fall, winter, spring, and summer were 7.4 ± 8.6, -0.5 ± 1.2, 1.1 ± 1.9, and 5.8 ± 2.7 mg m⁻²h⁻¹, respectively. Ammonia emission rates from flushed manure for fall, winter, spring, and summer were 3.9 ± 4.2, -0.5 ± 0.9, 0.8 ± 1.4, and 4.4 ± 1.2 mg m⁻²h⁻¹, respectively. Seasonal changes of air temperature and manure pH were key factors affecting ammonia emissions from manure storage in this study. Seasonal climate conditions including precipitations (rainstorms and snows) and icing can cause reduction of ammonia emissions from manure storage in open air. More attention should be paid to reduce ammonia emissions in warmer seasons, e.g., by covering the storage facilities.
- Assessing the Potential Use of Teff as an Alternative Grain Crop in VirginiaColeman, Jennifer Marie (Virginia Tech, 2012-04-27)Teff (Eragrostis tef (Zucc.)) is an annual, warm-season cereal crop most notable for its gluten-free, nutrient-packed seed. Experiments were conducted in two regions of Virginia (Blacksburg and Steeles Tavern) in 2010 and 2011 to determine the grain production potential of two teff varieties (brown and white). Additionally, commercially purchased teff flour was evaluated for its suitability in producing a satisfactory baked product. Teff varieties were planted in early June and July at a seeding rate of 6 kg PLS ha??. Nitrogen fertilizer was applied at planting in the form of urea at a rate of 56 kg ha??. The experimental design was a randomized complete block with a two-way factorial treatment structure (variety and planting date) and four replications. Grain yield and nutritive value, straw yield and quality, and plant height were evaluated for each variety and planting date at Steeles Tavern in 2010. Due to failure in crop establishment and difficulties involved in threshing and processing the harvested crop, no data is available in 2010 or 2011 for Kentland or in 2011 for Steeles Tavern. In 2010 at Steeles Tavern, grain yield was significantly higher for the brown variety (367 kg ha??) compared to the white variety (97 kg ha??) for both planting dates. There was no significant difference in straw yield between varieties or planting dates with straw yield averaging 2645 and 2475 kg DM ha?? for brown and white varieties, respectively. Precipitation accumulation at Steeles Tavern was higher in 2010 (greater than 10 cm) during June and July compared to 2011 and the historic average. This may explain why the plots in 2010 were able to successfully establish and out compete weeds. In the lab, four types of baked products were tested to determine the suitability of teff for baked goods. Cakes, cookies, biscuits and bread were tested with varying treatments of teff: control (100% wheat flour) and 10, 20, 30, 40 and 100% teff flour. Each treatment was replicated three times for each product. Generally, bread and cake volumes decreased as the percent of teff increased. Teff flour was best suited for use in cookie and biscuit products compared to cakes and breads since cookies and biscuits require less leavening. Overall, both experiments (field and laboratory) demonstrated the potential of teff as an alternative grain crop in Virginia. However, additional research is needed to overcome problems associated with establishment, harvest, threshing and processing.

Oils produced from algae, fungi, bacteria, molds, and yeast can also be used to produce biodiesel. The production and use of biodiesel is increasing across the United States. Commonly cited advantages of using biodiesel include: a good substitute for petroleum diesel fuel; using up to a 20% blend in most diesel equipment with no/minor modifications; burns clean, resulting in reduced toxic tailpipe emissions and gases associated with global warming; nontoxic, biodegradable, and suitable for use in sensitive environments; provides an additional market for vegetable oils and animal fats; provides farmers with the ability to grow their own fuel; and decreases U.S. dependence on imported oil if fuel feedstocks are grown locally. This publication addresses facts about using biodiesel fuel and presents the basic properties of biodiesel fuel.

- Characterization of a Novel Biodegradable Material to Reduce Emission of AmmoniaAdjei, Thomas (Virginia Tech, 2007-12-06)A novel biodegradable ammonia control material was developed from steam exploded corn cob and its adsorption capacity was studied by packed column and titration method. The packed column studies showed that the maximum absorption capacities of the raw corn cob (RCC) and the steam exploded corn cob (SECC) were 10.45 mg NHB3B/gRCC and 59.80 mg NHB3B/gSECC respectively. However, the titration of the water slurries with a NHB4BOH showed that the capacity of the SECC was 14.4 times that of RCC. The large difference between the packed column (SECC/RCC = 5.7) and the slurry titration (SECC/RCC = 14.4) was probably because: (1) the initial ammonia reaction products blocked the pores of the SECC and reduced diffusion into the pore structure; (2) the ammonia gas flow rates were too high and therefore the gas did not penetrate the pores; (3) the gas contact time was below the equilibrium value; and (4) since interior pore surface area is usually larger than the external surface area of a particle, it appears the low column SECC/RCC ratio is due to reactions on the SECC particle surface whereas the slurry result was a combination of both. Fourier Transform Infrared, FTIR spectroscopy was conducted on RCC, SECC, ammonia adsorbed on RCC and ammonia adsorbed on SECC in the range 4000–700 cmP-1P. The FTIR bands in the region between 1500 and 2000 cmPâ 1P showed a considerable difference between RCC and SECC. When SECC was treated with ammonia, the carboxylic functional group peak at 1700 cmP-1P was reduced and a new peak was observed at 1584 cmP-1P. The adsorption, desorption test and the heat of adsorption results suggested combined physisorption and chemisorption of ammonia on SECC but chemisorption was found to play an important role in ammonia removal. The BET specific surface area of RCC was 3.4 m2/g whilst that SECC was less than 1 m2/g. Although SECC had a low surface area compared with RCC its adsorption capacity was found to be greater than that of RCC meaning the adsorption process is chemically controlled. Also, the pore size distribution showed that RCC exhibited both macroporosity and mesoporosity whilst SECC showed only mesoporosity. It is interesting to note that upon steam exploding RCC, the macropores within RCC collapsed to form more mesopores in SECC. The high uptake of SECC was determined to be its small pore width compared to that RCC. Simultaneous Differential Scanning Calorimetry, DSC and Thermal Gravimetric Analyzer, TGA, was used to determine the heat of adsorption of ammonia on SECC. The heat of adsorption of ammonia on SECC was 33.00 kJ per mole of NHB3B. This study shows that SECC could be potentially used to remove NHB3B from various emission sources.
- Comparison of Dairy Manure Anaerobic Digestion Performance in Gas-lift and Bubble Column DigestersTang, Jing (Virginia Tech, 2009-12-04)Anaerobic digestion is one of the most promising management options for dairy manure treatment. Manure wastewater from dairy farms has been used for methane production for decades. However, performance failure due to inadequate mixing is routine. In general, the mixng of anaerobic digester is achived throguh mechnical stirring, liquid circulation, and gas circulation, among which the gas circulation proves to be the most effcient way. In this work, we studied the liquid flow pattern of two differetn type of gas-circualtion based anaerobic digesters, with the aim to understand the effects of hydrodynamic behavior of the digesting liquid on the anaerobic digestion performance, so a better mixing strategy can be provided. We used two 20-L gas circulation based anaerobic digesters with confined (gas-lift) and unconfined (bubble column) design. The anaerobic digestion performance and mixing behaviors were studied at different gas recirculation rate. It was found that the biogas production from the bubble column was constantly higher than that from gas-lift digester. However, the overall flow of the two digesters, which is indicated by residence time distribution (RTD), showed a similar pattern. Furhter investigation of local liquid flow behavior using Computational Fluid Dynamic (CFD) indicate that the bubble column accumulated higher portion of sludge in the bottom of the digester, which has a higher TS and VS, COD, and biomethane production potential than those from the gas-lift digester. This is the reason that the biogas production from the bubble column is higher than the gas-lift digester. Through this study, a thorough characterization of the flow behavior of the anaerobic digester were developed, and provided a deep insight of its influence on the anaerobic digestion performance.
- Compartmental Process-based Model for Estimating Ammonia Emission from Stored Scraped Liquid Dairy ManureKarunarathne, Sampath Ashoka (Virginia Tech, 2017-07-06)The biogeochemical processes responsible for production and emission of ammonia from stored liquid dairy manure are governed by environmental factors (e.g. manure temperature, moisture) and manure characteristics (e.g. total ammoniacal nitrogen concentration, pH). These environmental factors and manure characteristics vary spatially as a result of spatially heterogeneous physical, chemical, and biological properties of manure. Existing process-based models used for estimating ammonia emission consider stored manure as a homogeneous system and do not consider these spatial variations leading to inaccurate estimations. In this study, a one-dimensional compartmental biogeochemical model was developed to (i) estimate spatial variation of temperature and substrate concentration (ii) estimate spatial variations and rates of biogeochemical processes, and (iii) estimate production and emission of ammonia from stored scraped liquid dairy manure. A one-dimension compartmentalized modeling approach was used whereby manure storage is partitioned into several sections in vertical domain assuming that the conditions are spatially uniform within the horizontal domain. Spatial variation of temperature and substrate concentration were estimated using established principles of heat and mass transfer. Pertinent biogeochemical processes were assigned to each compartment to estimate the production and emission of ammonia. Model performance was conducted using experimental data obtained from National Air Emissions Monitoring Study conducted by the United States Environmental Protection Agency. A sensitivity analysis was performed and air temperature, manure pH, wind speed, and manure total ammoniacal nitrogen concentration were identified as the most sensitive model inputs. The model was used to estimate ammonia emission from a liquid dairy manure storage of a dairy farm located in Rockingham and Franklin counties in Virginia. Ammonia emission was estimated under different management and weather scenarios: two different manure storage periods from November to April and May to October using historical weather data of the two counties. Results suggest greater ammonia emissions and manure nitrogen loss for the manure storage period in warm season from May to October compared to the storage period in cold season from November to April.

- Conservation of Nitrogen via Nitrification and Chemical Phosphorus Removal for Liquid Dairy ManureDeBusk, Jo (Virginia Tech, 2007-11-27)The objectives of this study were to (1) determine an intermittent aeration strategy that could be used to conserve nitrogen (N) via nitrification in dairy manure, (2) determine the effect of recycled flush water on the bio-availability of N during nitrification, and (3) determine effective and economical dosages of chemicals to remove phosphorus (P) from liquid dairy manure. Intermittent aeration strategies, defined in terms of time the aerator is on and off (ON h:OFF h), could be used to conserve N in dairy manure. Testing of four treatments (continuous aeration [100%], 1h:0.33h [75%], 1h:0.67h [60%], and 1h:1h [50%]) showed that only treatments using air provided for 100% and 75% of the time could support nitrification. The 100% and 75% aeration treatments conserved an average of 38% and 25% of influent total ammonia nitrogen (TAN) as nitrite-N+nitrate-N, respectively. Less than 2% of influent TAN was conserved using 60% and 50% treatments. The effect of manure handling technique on N bioavailability and nitrification was tested using flushed and scraped dairy manure. Nitrification was inhibited in scraped manure. Four aluminum- and iron-based salts and five cationic polyacrylamide polymers were evaluated for P removal using jar tests. Ferric chloride (FeCl3·6H2O), aluminum sulfate (Al2[SO4]3·13H2O, alum), and Superfloc 4512 were selected for further study. Polymer addition enhanced floc size and improved P removal. Treatment of manure (0.89% total solids) from Tank 2 at Virginia Tech's dairy using either FeCl3 or alum in combination with polymer resulted in more than 90% P removal. Chemical treatment and transport of P-rich sludge from a 2,270 cubic meter storage tank would result in an estimated 40% cost savings over transport of the entire manure volume offsite for land application elsewhere. The manure treatment strategies tested provide some solutions to dairy farmers regarding adjustment of N:P ratios so that manure can be applied to meet nutrient needs of crops while adhering to regulations set forth by nutrient management plans.
- Discovery of a Novel Microalgal Strain Scenedesmus Sp. A6 and Exploration of Its Potential as a Microbial Cell FactoryGuimaraes Braga da Silva, Pedro Ivo (Virginia Tech, 2018-08-14)Microalgae are photosynthetic organisms considered to be one of the most promising high-value chemicals and biofuel-producing organisms. However, there are several challenges for the widespread implementation of industrial processes using microalgae. The work presented in this dissertation proposes solutions to the different challenges involving the use of microalgae as microbial cell factories. To investigate the application of anaerobic digestion as a way to generate nutrients for microbial growth, salmon offal was used as substrate for anaerobic digestion, and soil from a flooded run-off pond on the Virginia Tech campus in Blacksburg, VA. A fast reduction in volatile solids and the short-chain fatty acid production profile is favorable for the growth of microalgae. A novel algae strain Scenedesmus sp. A6 was isolated from a decorative waterfountain in a hotel in Madison, IN. Mixotrophic growth trials were conducted using wastewater from the salmon offal digestion, that demostrated the A6 isolate grows six times faster in the wastewater then autotrophically. Bioassays of ethanolic cell extracts of A6 cultures demonstrated antimicrobial activity against E. coli cells at concentrations above 50 µg/ml. Genome sequencing and assembly revealed multiple copies of genes involved with acetate and ammonia metabolism, and several genes involved with secondary metabolite synthesis. An alternative to the high capital investment of photobioreactors for the cultivation of microalgae is the use of open-source and open-hardware bioreactor controller. Here, the concept of an open-hardwate bioreactor control called ``BioBrain'' is introduced. The BioBrain device is based on the Arduino Mega micro-controller board, and is capable of monitoring and controlling culture conditions during simple strain characterization studies, with an estimated construction cost of less than $800 USD. Finally, a new primer design tool for the ligation-independant cloning technique 𝜆-PCR was developed called lambdaPrimeR. The contributions of this work are the discovery and development of different tools that can overcome the challenges of the use of microalgae as microbial cell factories in industrial processes.
- The Effect of a Trace Element Supplement on the Biomethane Potential of Food Waste Anaerobic DigestionGraff, Kelly Mackenzie (Virginia Tech, 2022-06-15)Food waste is a desirable feedstock for anaerobic digestion because it is high in moisture and is an easily degradable material. However, mono-digestion of food waste often fails due to the accumulation of volatile fatty acids. Supplementing trace elements is one strategy to combat this issue. This study examined the effect of supplementing trace elements (iron, nickel, selenium, molybdenum, magnesium, zinc, calcium, copper, manganese, cobalt) on the methane yield and organic waste destruction of anaerobically digested food waste. Methane yield of food waste with and without the inorganic salt trace element was determined by the gas density-based biomethane potential method at mesophilic (37°C) conditions over 30 days. The three treatments were inoculum only, food waste and inoculum, and food waste and inoculum with an added trace element solution. There was no significant difference between treatments in terms of waste stabilization (percent volatile solids, total solids, and total chemical oxygen demand reduction) between treatments. The average cumulative biogas produced was 41% higher, and the average total cumulative methane produced was 23% higher in the treatment with the trace element supplement. Mean methane yield was not different (p > 0.05) between treatments over the 30 days, and there was no difference (p > 0.05) in biomethane potential between treatments. In addition, greenhouse gas reduction potential was estimated from food waste streams in Montgomery, VA using anaerobic digestion. The purpose of this work was to (1) estimate the total mass of food waste produced in Montgomery, VA in a year, (2) use the results from the biomethane potential analyses to inform the sizing of a theoretical community digester in Montgomery, VA, and (3) estimate the greenhouse gas reduction potential of anaerobically digesting the food waste instead of sending it to landfill. Greenhouse gas reduction was calculated using the Climate Action Reserve Organic Waste Digestion Project Protocol guidelines. The greenhouse gas reduction potential was estimated as 6,532 tonnes of carbon dioxide equivalent per year (tCO2e/year), with approximately 693 m3 methane produced per day. In one year, the digester would generate an estimated 7370 kWh of energy which has the potential to power 149 homes for a year in Montgomery, VA. In addition, 4130 tonnes/year of composted digestate would be available as fertilizer for surrounding farms.
- The Effect of Thermophilic Anaerobic Digestion on Ceftiofur and Antibiotic Resistant Gene Concentrations in Dairy ManureHowes, Sasha Alyse (Virginia Tech, 2017-07-06)The prevalence of antibiotics on farms for therapeutic and prophylactic use in animals can cause negative effects on biomethane production during anaerobic digestion. Previous literature has found decreased biomethane production rates from a variety of antibiotics, but biogas inhibition differs between studies of continuous and batch reactors and the type of antibiotic studied. Cephalosporin drugs are the most common antibiotic class used to treat mastitis in dairy cows and can retain most of their bioactivity after excretion. Ceftiofur is a commonly used cephalosporin drug but no previous study investigating the effect of Ceftiofur on biomethane during continuous anaerobic digestion has been performed. The aim of this study was to examine the effect on biomethane production when manure from cows treated with Ceftiofur was anaerobically digested. Laboratory sized anaerobic digesters (AD) were run at thermophilic (55°C) temperatures and a 10 day hydraulic retention time. Manure from cows treated with Ceftiofur were fed to the antibiotic treatment reactors for 50 days. The reactor performance was measured by i) biomethane production, ii) waste stabilization in terms of solids and chemical oxygen demand, iii) change in mass of Ceftiofur and iv) change in concentration of antibiotic resistant genes, specifically cfx(A), mef(A), and tet(Q). There was statistically significant decrease in cumulative gas production due to the addition of Ceftiofur into the reactors, but no significant difference between treatments in waste stabilization in terms of percent volatile solids (VS) and total chemical oxygen demand (TCOD) reduction. Anaerobic digestion decreased the amount of Ceftiofur in manure, and the amount of Ceftiofur in the reactors reduced over the time of the experiment. Change in antibiotic resistant genes (ARGs) were gene dependent over time. Concentrations of tet(Q) reduced significantly between feed and effluent of both treatments, and cfx(A) reduced significantly for the control treatment but not the Ceftiofur treatment. Concentrations of mef(A) increased over time in both treatments. Overall, the addition of Ceftiofur in continuously operated anaerobic digesters negatively affected biomethane production, a value-added product responsible for on-farm renewable energy. However, anaerobic digestion does decrease the mass of Ceftiofur within manure, thereby reducing the environmental loading from run-off from farms.
- The effects of dietary protein content and manure handling technique on ammonia emissions during short-term storage of dairy cow manureSparks, J. (American Society of Agricultural and Biological Engineers, 2011)An improved understanding of the potential for dietary protein manipulation to reduce ammonia emissions from dairy farms during various stages of manure handling is needed for both modeling and policy-making efforts. The objective of this study was to assess the effects of dietary protein manipulation on ammonia emissions from relatively freshly voided dairy cow manure in three types of removal systems: scraped manure removal systems, flushed manure removal systems, and flushed manure removal systems with a solids separator. Emissions were measured using a dynamic flux chamber for 12 h or more. Ammonia fluxes and emission factors per mass of manure were not affected by dietary protein content because fluxes depended mainly on total ammoniacal nitrogen (TAN) concentration, which did not vary with diet. However, emissions on a per-cow basis were 12% lower with the diet containing 15.0% crude protein as compared to the one with 17.8% (a change in crude protein of 16%) due to reduced urine output. The largest absolute impact of dietary protein manipulation would be with separated liquids because their emission factor was approximately four times higher than for the other types of manure. While dietary protein manipulation can reduce ammonia emissions from manure during long-term storage, its effectiveness in the hours immediately after manure is excreted is limited because emissions are more sensitive to other factors, including temperature and extent of mixing of the manure, that vary widely under real operating conditions on a farm.
- The Effects of Manure Handling and Dietary Protein on Ammonia Fluxes from a Flush DairySparks, Jeffrey Allen (Virginia Tech, 2008-04-28)Atmospheric ammonia (NH₃(g)) poses an environmental concern mainly due its ability to form fine particulate matter (PM2.5 with a diameter smaller than 2.5 micrometers) in the form of ammonium nitrate (NH₄NO₃) and ammonium sulfate ([NH₄]₂SO₄). These forms of PM2.5 may reduce visibility, contribute to eutrophication through deposition, and be detrimental to human health through inhalation. A more complete understanding of ammonia emissions may bring significant PM2.5 reductions within grasp. According to recent studies, one of the largest contributors to atmospheric ammonia is dairy cattle excreta. In this study, seven lactating Holstein cows were subjected to four feeding trials with diets containing 14.5, 15.5, 17, and 18% crude protein (CP). The first objective was to determine the effect of protein content/intake on ammonia fluxes to the atmosphere from each stage of manure handling at a flush dairy. The second objective was to examine the effect of manure handling itself on ammonia fluxes. We examined four different stages of manure handling: 1. Fresh mixture of urine and feces to represent what may be scraped from a barn floor 2. Diluted mixture of urine and feces to represent what is flushed from a barn floor 3. Solids portion of a diluted mixture of urine and feces to represent what is retained by a solids separator at a dairy 4. Liquid portion of a diluted mixture of urine and feces to represent the permeate from a solids separator at a dairy Ammonia fluxes from scraped manure began low and rose to higher fluxes and peaked at an average of 25 hours after mixing urine and feces. Fluxes from flushed manure showed the same behavior. The average pH values for the scraped and flushed manure were relatively low at 6.87±0.17 and 7.05±0.13, respectively, and unfavorable toward ammonia versus ammonium. Fluxes from the separated solids and liquid manure, whose pH values were higher at 8.70±0.30 and 8.55±0.19, respectively, peaked almost immediately. For a time period of 18 hours after beginning flux measurements from the separated liquid manure, fluxes stayed relatively constant. Compared to hours 0-9, fluxes from the separated solids manure decreased 11% in hours 9-18. These results indicate that ammonia fluxes from the scraped and flushed manure were initially limited by the relatively low pH of the slurries, until they aged and released carbon dioxide, after which fluxes became controlled by temperature. Fluxes from the separated solids and liquid manure, however, were temperature-controlled from the start, as this manure had already undergone some aging and the initial pH was more favorable to ammonia loss. Multivariable regressions predicting the mass of ammonia lost to the atmosphere as a function of total ammoniacal nitrogen (TAN) and temperature (T) show that fluxes from the scraped and flushed manure are very sensitive to temperature compared to the separated solids and liquid manure. The same regressions show that ammonia fluxes from the separated solids and liquid manure are predicted well by TAN and T (R² = 0.906 and 0.812, respectively), indicating that dietary protein manipulation, which affects TAN content of the manure, may have a greater effect in this stage of handling. Regressions predicting the mass of ammonia lost to the atmosphere as a function of crude protein intake show crude protein intake is a poor predictor of the overall ammonia flux from a flush dairy. Low correlation coefficients from these regressions are likely a result of cow-to-cow variability in the effect of crude protein intake on ammonia emissions. Significant reductions in ammonia fluxes are possible; however, they will require new regulations on manure handling at flush dairies or incentives for flush dairies to pursue alternate means of manure handling with low emissions. Dairies can begin attacking the problem of excessive emissions by lowering the dietary protein content for older cows to reduce the mass of TAN excreted in their manure, thereby reducing ammonia fluxes in accordance with Henry's law. Once manure is excreted on the barn floor, dairies must encourage mixing between urine and feces and use the natural acidity of the feces in delaying emissions by converting aqueous ammonia in the urine to ammonium. Dairies can achieve well-mixed slurries by manually mixing the manure or with a sloped floor, channeling urine and feces to a common area. Covering holding tanks containing flushed manure to minimize CO₂ stripping and a rise in pH is one measure that may reduce ammonia fluxes. Finally, requiring nitrification or nitrification and denitrification in on-site storage tanks will biologically convert ammonia to nitrate or nitrogen gas, respectively, reducing fluxes from the separated liquid manure. Although burdensome to dairies, these measures would drastically reduce ammonia emissions from flush dairies. Regulatory agencies may choose to ignore the separated solids manure when considering strategies to reduce ammonia emissions, since ammonia volatilization becomes limited by aqueous ammonia in the first 18 hours following separation.
- Energy-efficient Wastewater Treatment by Microbial Fuel Cells: Scaling Up and OptimizationGe, Zheng (Virginia Tech, 2015-11-06)Microbial fuel cells (MFCs) are potentially advantageous as an energy-efficient approach to wastewater treatment. For single-chamber tubular MFCs, anode effluent is used as catholyte instead of tap water or buffer solutions. Therefore, exposing cathode electrode to atmosphere could be also considered as a passive aeration for further aerobic oxidation of organics and nitrification. Based on several bench-scale studies, a 200-L scale MFC system with passive aeration process has been developed for treating actual municipal wastewater after primary clarification. The integrated system was able to remove over 80% organic contaminants and solid content from primary effluent. Through parallel and serial electricity connection, the power output of ~200 mW and the conversion efficiency of ~80% for charging capacitors were achieved by using commercially available energy harvesting device (BQ 25504). The treatment system is energy-efficient for the energy saving from aeration and sludge treatment while partial energy recovery as direct electricity can be utilized on site to power small electric devices. However, the post treatments are required to polish the effluent for nutrients removal.
