(Elsevier, 2020-05-01)Goblet cells secrete mucin 2 (Muc2), which is a major component of the mucus that lines the intestinal tract and creates a protective barrier between pathogens and the intestinal epithelial cells and thus are important for chick health. The objectives of this study were to determine the age-specific and intestinal segment–specific expression of Muc2 mRNA and changes in the number of goblet cells from late embryogenesis to early after hatch. Small intestinal samples from the duodenum, jejunum, and ileum were collected from Cobb 500 broilers at embryonic day 19 (e19), day of hatch (doh), and day 2 and 4 after hatch. Cells expressing Muc2 mRNA and mucin glycoprotein were detected by in situ hybridization or alcian blue and periodic acid–Schiff staining, respectively. Along the villi, there were many more cells expressing Muc2 mRNA than those stained for mucin glycoprotein. In the crypt, cells expressing Muc2 mRNA did not stain for mucin glycoprotein. There was an increase in the density of goblet cells in the villi and Muc2 mRNA expressing cells in the crypts of the jejunum and ileum from e19 to doh and day 2 to day 4, with no change between doh and day 2. In contrast, in the duodenum, the density of goblet cells in the villi and Muc2 mRNA expressing cells in the crypts remained constant from e19 to day 4. At day 4, the villi in the ileum had a greater density of goblet cells than the duodenum. In the crypt, the ileum had a greater density of Muc2 mRNA expressing cells than the duodenum at doh, and the ileum and jejunum both had greater densities of Muc2 mRNA expressing cells than the duodenum at day 4. These results indicate that the population of goblet cells has reached a steady state by doh in the duodenum, whereas in the jejunum and ileum, a steady-state population was not reached until after hatch.
- Effects of available phosphorus source and concentration on performance and expression of sodium phosphate type IIb cotransporter, vitamin D-1 alpha-hydroxylase, and vitamin D-24-hydroxylase mRNA in broiler chicksOmara, Islam I.; Mou, C. T.; Persia, Michael E.; Wong, Eric A. (Elsevier, 2020-04-01)This experiment was conducted to examine the effect of 2 phosphorus (P) sources on broiler performance to day 14. The P bioavailability was estimated using bird performance and tibia ash measurements, whereas P digestibility, intestinal P transporter, kidney vitamin D-1α-hydroxylase, and vitamin D-24-hydroxylase mRNA abundances were also determined. Slope regression analysis was used to determine the bioavailability of dicalcium phosphate (Dical P) and nanocalcium phosphate (Nano P) with dietary available P (AvP) set to 0.20% P (control) using AvP from the major ingredients and Dical P. The experimental treatments were achieved by supplementation with either Dical P or Nano P to generate 0.24, 0.28, 0.32, and 0.36% AvP. A total of 648-day-old unsexed broiler chicks were divided into 72 birds per treatment (8 replicate cages of 9 birds). Slope regression analysis showed positive linear relationships between BW, feed intake (FI), tibia ash weight (TAW), and tibia ash percentage (TAP) with dietary Dical P and Nano P levels. Comparisons between regression slopes for Dical P and Nano P fed birds were not significantly different for BW, feed intake, tibia ash weight, and tibia ash percentage, indicating similar P bioavailability from Dical P and Nano P. There were interactions between P source and AvP for feed efficiency (FE) and apparent ileal P digestibility (AIPD). Dicalcium phosphate had greater FE than Nano P at 0.28% AvP and greater AIPD than Nano P at 0.24% AvP. The addition of AvP from Dical P and Nano P resulted in reduced sodium phosphate cotransporter mRNA abundance in the duodenum in a dose–dependent response. In the kidney, vitamin D-1α-hydroxylase mRNA abundance was greater at 0.36% Nano P compared with control, but there was no difference with Dical P. There was no difference in vitamin D-24-hydroxylase mRNA abundance between control and supplementation with Nano P or Dical P. In conclusion, Nano P and Dical P had the same bioavailability but had different effects on gene expression.
- Glucose Tolerance and Plasma Non-Esterified Fatty Acid Levels in Chickens Selected for Low Body Weight, Red Junglefowl, and their Reciprocal CrossSutherland, Dez-Ann A. (Japan Poultry Science Association, 2019-10-01)Responses of an individual to food deprivation, such as a 16-h fast, are complex, and are influenced by environmental and genetic factors. Domestication is an ongoing process during which adaptations to changing environments occur over generations. Food deprivation by their caretakers is less for domestic chickens than for their junglefowl ancestors. Unlike domestic chicken, the junglefowl adapted over generations to periods of food deprivation, which may be reflected in differences in metabolic responses to brief periods without food. Here, we compared the blood glucose and plasma levels of non-esterified fatty acids (NEFA) among four populations when deprived of feed for 16 h. The four populations included a domestic White Rock experimental line (LWS) maintained for generations under ad libitum feeding, adult red junglefowl (RJF), and a reciprocal cross of the lines. Although there were significant differences in adult (31-week) body weight between the RJF (683 g) and LWS (1282 g), with the weight of F1 crosses being intermediate, the amount of abdominal fat relative to body weight was similar for all populations. Patterns for blood glucose responses to a glucose bolus after a 16-h fast were similar for the initial and final points in the parental and cross populations. However, RJF reached their peak faster than LWS, with the reciprocal cross intermediate to the parental populations. Plasma NEFA concentrations were higher after the 16-h fast than in fed states, with no population differences for the fasting state. However, in the fed state, NEFA levels were lesser for LWS than for others, which was reflected further in percentage change from fed to fasted. This larger change in LWS suggests differences in mobilization of energy substrates and implies that during domestication or development of the LWS line, thresholds for responses to acute stressors may have increased.
- Impact of Water Sanitation on Broiler Chicken Production and Welfare ParametersJacobs, Leonie; Persia, Michael E.; Siman-Tov, Nisana; McCoy, Jesse; Ahmad, Mueez; Lyman, Joe; Good, Lindsey (Elsevier, 2020-03)The objective of the study was to assess the impact of drinking water sanitation on performance and welfare outcomes in broiler chickens kept on fresh or used litter. We hypothesized that under a more challenging environment (used litter), the impact of water sanitation would differ to that in a less challenging environment. Seventy-two pens housed 1,944 male broilers for 35 D. The treatments included a tap water control, water with fully activated ClO2, water with partially activated ClO2, and water with iodine, offered to birds housed on either clean or used pine shavings. The latter resulted from housing 20 birds per pen that had been vaccinated with Coccivac-B. The intended setup was successful as birds on used litter showed worsened feed conversion, body weights, and daily gains early in the production cycle, although these differences were lost over time. Water sanitation did result in altered drinking behavior, with fewer drinking bouts for birds on iodine treatment than for birds on control and fully activated ClO2 treatments. Iodine-treated birds showed less severe footpad lesions than birds on other treatments. Similarly, birds on fresh litter had smaller lesions than those on used litter. Iodine- and fully activated ClO2–treated birds experienced less chronic stress than control birds. The impact of water sanitation did not differ when birds were in a challenging environment (used litter) compared with those in a cleaner environment. Water sanitation did provide some benefits for production (body weight and daily gain) and welfare outcomes (footpad dermatitis and chronic stress).
- The inhibitory effect of trans-10,cis-12 conjugated linoleic acid on sterol regulatory element binding protein-1 activation in bovine mammary epithelial cells involved reduced proteasomal degradation of insulin-induced gene-1Chen, Liang; Lengi, Andrea J.; Corl, Benjamin A. (American Dairy Science Association, 2021-07-15)Trans 10,cis-12 conjugated linoleic acid (t10,c12 CLA) is well recognized as a key CLA isomer responsible for the reduction in milk fat synthesis that leads to milk fat depression in dairy cows. Sterol regulatory element binding protein-1 (SREBP1) is a key transcription factor in bovine mammary gland coordinating transcription of the genes for fatty acid synthesis. SREBP1 activation requires the removal of insulin-induced gene-1 (Insig1) that serves as a repressor of SREBP1 in the endoplasmic reticulum (ER). We hypothesized that t10,c12 CLA reduced SREBP1 activation by delaying Insig1 degradation. In the present study, we used undifferentiated bovine mammary epithelial cells (MAC-T cells) and treated them with t10,c12 CLA for 6 h. We found that SREBP1 protein expression declined over 56% when cells were treated with 60 µM or greater concentration of t10,c12 CLA. Such inhibitory effects were also observed in the mRNA expression of SREBP1-regulated genes including SREBP1, fatty acid synthetase, stearoyl-CoA desaturase, and Insig1. Compared with no CLA group, 60 µM or higher concentration of t10,c12 CLA increased Insig1 protein expression over 2-fold in cells transfected with FLAG-tagged Insig1. This stimulatory effect was not specific to t10,c12 CLA but also other polyunsaturated fatty acids including cis-9,trans-11 CLA and linoleic acid. Oleic acid had no effect on Insig1 protein expression, whereas palmitic acid decreased Insig1 protein expression. Further investigation revealed that increased abundance of FLAG-Insig1 with t10,c12 CLA was due to the inhibition of the proteasomal degradation of Insig1. The t10,c12 CLA delayed the Insig1 decay when protein synthesis was blocked. Immunoprecipitation also confirmed that the interaction between ubiquitin-like domain-containing protein 8 and Insig1, the key step of removing Insig1 from ER and freeing SREBP1 for proteolytic processing, was inhibited by t10,c12 CLA, but not palmitic acid. These findings suggested that t10,c12 CLA played a role in regulating SREBP1 activation by reducing proteasomal degradation of Insig1. We concluded that stabilized Insig1 retained SREBP1 in the ER from activation, thus reducing lipogenic gene transcription.
- Intermittent Bolus Compared with Continuous Feeding Enhances Insulin and Amino Acid Signaling to Translation Initiation in Skeletal Muscle of Neonatal PigsSuryawan, Agus; El-Kadi, Samer Wassim; Nguyen, Hanh V.; Fiorotto, Marta L.; Davis, Teresa A. (Oxford University Press, 2021-09-01)Background: Nutrition administered as intermittent bolus feeds rather than continuously promotes greater protein synthesis rates in skeletal muscle and enhances lean growth in a neonatal piglet model. The molecular mechanisms responsible remain unclear. Objectives: We aimed to identify the insulin- and/or amino acid-signaling components involved in the enhanced stimulation of skeletal muscle by intermittent bolus compared to continuous feeding in neonatal pigs born at term. Methods: Term piglets (2-3 days old) were fed equal amounts of sow milk replacer [12.8 g protein and 155 kcal/(kg body weight · d)] by orogastric tube as intermittent bolus meals every 4 hours (INT) or by continuous infusion (CTS). After 21 days, gastrocnemius muscle samples were collected from CTS, INT-0 (before a meal), and INT-60 (60 minutes after a meal) groups (n = 6/group). Insulin- and amino acid-signaling components relevant to mechanistic target of rapamycin complex (mTORC) 1 activation and protein translation were measured. Results: Phosphorylation of the insulin receptor, IRS-1, PDK1, mTORC2, pan-Akt, Akt1, Akt2, and TSC2 was 106% to 273% higher in the skeletal muscle of INT-60 piglets than in INT-0 and CTS piglets (P < 0.05), but phosphorylation of PTEN, PP2A, Akt3, ERK1/2, and AMPK did not differ among groups, nor did abundances of PHLPP, SHIP2, and Ubl4A. The association of GATOR2 with Sestrin1/2, but not CASTOR1, was 51% to 52% lower in INT-60 piglets than in INT-0 and CTS piglets (P < 0.05), but the abundances of SLC7A5/LAT1, SLC38A2/SNAT2, SLC38A9, Lamtor1/2, and V-ATPase did not differ. Associations of mTOR with RagA, RagC, and Rheb and phosphorylation of S6K1 and 4EBP1, but not eIF2α and eEF2, were 101% to 176% higher in INT-60 piglets than in INT-0 and CTS piglets (P < 0.05). Conclusions: The enhanced rates of muscle protein synthesis and growth with intermittent bolus compared to continuous feeding in a neonatal piglet model can be explained by enhanced activation of both the insulin- and amino acid-signaling pathways that regulate translation initiation.
- Lysine and sulfur amino acid requirements of broiler chicks over short time periods within the starter phaseSix experiments were conducted to validate the hypothesis that Lys and SAA requirements decrease within the starter phase using 3-d periods from 2 to 11 d of age. In the first 3 experiments, 7 diets were generated by adding L-Lys to a lysine-deficient basal diet in 0.10% increments, ranging from 0.85 to 1.45% digestible Lys. In experiments 4 to 6, and 7 diets were generated by adding 0.07% increments of DL-methionine to a SAA-deficient diet to produce diets ranging from 0.63 to 1.04% DSAA. The linear broken line estimate for digestible Lys was 1.22, 1.17, and 1.16% for BWG and 1.31, 1.21, and 1.14% for FCR in experiments 1, 2, and 3, respectively. The linear broken line estimate for DSAA was 0.82, 0.81, and 0.94% for BWG and 0.82, 0.80, and 0.90 for FCR in experiments 4, 5, and 6, respectively. These results indicated that Lys requirements decreased linearly as hypothesized, however, the SAA requirements did not follow same pattern.
- Northern fowl mite infestation affects the nocturnal behavior of laying hensJacobs, Leonie; Vezzoli, Giuseppe; Beerda, Bonne; Mench, Joy A. (Elsevier, 2019-07-01)The northern fowl mite (Ornithonyssus sylviarum) is a key pest for caged laying hens. High infestation levels can lead to anemia, reduced production, and mortality. Although the mites cause skin irritation and thus likely affect resting behavior, the nocturnal behavior of infested chickens has not yet been studied. We investigated the impact of mite infestation on nighttime behavior of 16 beak-trimmed White Leghorn hens. The hens were housed individually in wire cages. They were experimentally inoculated with approximately 35 mites at 25 weeks of age and observed for the behaviors dozing, sleeping, preening and being active. Continuous observations were made from video recordings taken from 22:00 h until 06:00 h for two consecutive nights at pre-infestation week 0 and post-infestation weeks 3, 5 and 7. Mite infestation levels were measured weekly on an 8-point scale (0 (no mites) to 7 (≥10,000 mites per hen)). For statistical analyses with linear mixed models, mite infestation levels were categorized as score 0 (no mites), 3–4 (51–500), 5 (501–1000) and 6–7 (>1000). Higher levels of mite infestation were associated with hens spending less time dozing (P < 0.001) and more time preening (P < 0.001) and being active (P = 0.012). Dozing decreased from 90% of the observed time for mite score 0 to 76% for score 6–7. Preening increased from 2% (score 0) to 9% (6–7) and time spent being active increased from 1% (0) to 7% (6–7). Mite infestation increased the number of uninterrupted bouts of all behaviors (P ≤ 0.020), especially dozing and preening (P < 0.001), suggesting restlessness in the mite-infested hens due to irritation and itching. The mite-infested hens’ increased nocturnal activity, including preening, as well as the fragmentation of behavioral activities together with decreased dozing, indicate disturbed resting behavior and suggest a reduction in the welfare of mite-infested hens.
- Remedying Contact Dermatitis in Broiler Chickens with Novel Flooring TreatmentsFreeman, Nathan; Tuyttens, Frank A. M.; Johnson, Alexa; Marshall, Victoria; Garmyn, An; Jacobs, Leonie (MDPI, 2020-10-01)Contact dermatitis (footpad dermatitis (FPD), hock burns, and breast dermatitis) is a welfare issue for broiler chickens, causing pain and behavioral restrictions. Once lesions develop, often nothing is done to remedy the issue for the affected flock. Our objective was to evaluate novel flooring treatments at the flock level by providing preventative and remedial treatments against contact dermatitis, plumage soiling, and gait impairment. Broilers (n = 546) were housed in 42 pens, with 13 birds/pen. The flooring treatments (four) included used litter (NEG), new pine shavings replaced regularly (POS), a mat filled with 1% povidone-iodine solution (MAT), and the iodine mat placed on a slatted floor (SLAT). Flooring treatments were provided from day one of age (preventative approach; PREV) or day 29 (remedial approach; REM). Contact dermatitis, soiling, gait, and weight were recorded weekly (seven birds/pen). Results showed a treatment effect for all measures, dependent on bird age. Overall, the POS treatment resulted in the best welfare outcomes (FPD, hock burns, and gait). The worst contact dermatitis was found in the MAT and SLAT groups. NEG birds showed little contact dermatitis, opposite to expectations. Weights were lower for PREV-POS in week seven only. The treatments with povidone-iodine were deemed ineffective against contact dermatitis. Access to clean litter prevented and remedied contact dermatitis, and a comparable approach may be commercially feasible.
- Research Note: Intestinal morphology and gene expression changes in broilers supplemented with lysolecithinCloft, Sara E.; Jia, Meiting; Wong, Eric A. (Elsevier, 2021-07-01)Lysolecithin is used as a feed additive to aid fat digestion and absorption in broiler chickens. Previous research has shown that dietary fat source influences how broilers respond to lysolecithin supplementation. Therefore, the objective of this study was to investigate the effect of lysolecithin on a diet formulated with soybean oil on jejunum morphology and expression of selected genes in broiler chickens. Male Cobb 500 chickens were fed a Control diet or the Control diet supplemented with lysolecithin (TRT) from day of hatch to day 28. Jejunal samples were collected at day 10 for morphological and gene expression analysis. Feeding the TRT diet did not affect BW, villus height (VH), crypt depth (CD) or VH/CD ratio compared to Control fed chickens. Differential gene expression in the jejunum was analyzed using a custom microarray. Using a t test, 36 genes were found to be upregulated in TRT fed chickens compared to chickens fed the Control diet. The two most upregulated genes were carbonic anhydrase VII and interleukin 8-like 2, which are associated with healthy intestines. In summary, lysolecithin supplementation in a diet formulated with soybean oil caused no morphological changes but upregulated a number of genes in the jejunum.
- Short-Term Adaptation of Dairy Cattle Production Parameters to Individualized Changes in Dietary Top DressPrice, Tanner P.; Souza, Vinicius C.; Liebe, Douglas M.; Elett, Mark D.; Davis, Ty C.; Gleason, Claire B.; Daniels, Kristy M.; White, Robin R. (MDPI, 2021-12-01)Immediate and short-term changes in diet composition can support individualized, real-time interventions in precision dairy production systems, and might increase feed efficiency (FE) of dairy cattle in the short-term. The objective of this study was to determine immediate and short-term effects of changes in diet composition on production parameters of dairy cattle fed varying amounts of top dressed commodities. A 4 × 4 replicated Latin square design was used to evaluate responses of twenty-four Holstein cows fed either no top dress (Control) or increasing amounts of: corn grain (CG), soybean meal (SBM), or chopped mixed grass hay (GH) top dressed on a total mixed ration (TMR) over four, 9-day periods. Throughout each period, top dressed commodities were incrementally increased, providing 0% to 20% of calculated net energy of lactation (NEL ) intake. Measured production responses were analyzed for each 9-d period using a mixed-effects model considering two different time ranges. Samples collected from d 3 and 4 and from d 7 and 8 of each period were averaged and used to reflect “immediate” vs. “short-term” responses, respectively. In the immediate response time frame, control fed cows had lower milk yield, milk fat yield, and milk true protein yield than CG and SBM supplemented animals but similar responses to GH supplemented animals. Milk fat and protein percentages were not affected by top dress type in the immediate term. In the short-term response time-frame, GH supplemented animals had lower DMI and milk fat yield than all other groups. Control and GH supplemented cows had lower milk yield than CG and SBM fed cows. In the immediate response time frame, FE of SBM supplemented cows was superior to other groups. In the short-term time frame, FE of GH and SBM groups was improved over the control group. Results suggest that lactating dairy cows show rapid performance responses to small (<20% NEL ) changes in dietary composition, which may be leveraged within automated precision feeding systems to optimize efficiency of production. Before this potential can be realized, further research is needed to examine integration of such strategies into automatic feeding systems and downstream impacts on individual animal FE and farm profitability.
- Uneven milking intervals are adequate to achieve the benefits of increased milking frequency in early lactationHanling, H. H.; McGilliard, M. L.; Corl, Benjamin A. (American Dairy Science Association, 2021-08-01)Increasing milking frequency (MF) increases milk yield (MY) and farm profit, and optimal milking intervals (MI) prevent milk production decline. The objective of this experiment was to compare the MY effect of even and uneven 4 times daily (4×) MI in early lactation under increased MF. Fourteen multiparous and 6 primiparous cows were milked using unilateral frequent milking, with right udder halves milked 4× and left udder halves milked 2 times daily (2×) for 20 d in early lactation starting on d 5 postpartum. Ten (7 multiparous and 3 primiparous) cows were allocated evenly based on parity and assigned to either the even or the uneven MI groups distinguished by intervals of 9:3:9:3 h or 6:6:6:6 h. The left and right udder halves were milked at 0100 and 1300 h. The right udder glands were additionally milked at 0400 and 1600 h for the uneven MI group and at 0700 and 1900 h for the even MI group. Milk from each udder half was weighed and sampled for components on the final day of treatment and at 60, 120, 180, 240, and 300 d in milk. The overall effect of 4× milking on the right udder halves was a 5.96 ± 0.70 kg/d increase in MY on d 21 of unilateral frequent milking compared with the 2× udder halves. This elevated MY continued through 300 d in milk and averaged 1.56 ± 0.70 kg/d. Increased MF in early lactation increased the udder half difference in total yield throughout a 300-d lactation by 508 kg for milk, 25 kg for milk fat, and 15 kg for milk protein. Increased MF in early lactation increased milk component yields, but there were no differences between MI groups. The lack of treatment difference may be beneficial to farmers. The ability to achieve the same increased MY effect with uneven MI may optimize labor efficiency because early-lactation cows could be milked at the beginning and end of milking sessions. Farmers would not have to add additional milking sessions to achieve the enhanced MY response regardless of normal milking session length.
- Venous blood gas and chemistry components are moderately heritable in commercial white egg-laying hens under acute or chronic heat exposureRowland, Kaylee; Persia, Michael E.; Rothschild, Max F.; Schmidt, Carl J.; Lamont, Susan J. (Oxford University Press, 2019-09-01)Heat stress has a large negative impact on poultry around the world in both intensive and small-scale production systems. Better understanding of genetic factors contributing to response to high ambient temperatures would provide a basis to develop strategies for alleviating negative impacts of heat on poultry production. The objective of this work was to characterize the genetic control (heritability estimate and quantitative trait loci (QTL)) of blood chemistry components before and after exposure to acute and chronic high ambient temperature in a commercial egg laying line Hy-Line W-36 female parent line mature hens were exposed to 4 wk of daily cyclic heat exposure. Blood was collected pre-heat, on the first day of heat, and 2 and 4 wk post heat initiation and analyzed immediately using an i-STAT® hand-held blood analyzer. Thirteen blood components were quantified at the 4 time points: pH, pCO2, pO2, HCO3, TCO2, sO2, iCa, Na, K, base excess, glucose, “hematocrit” (estimated from blood electrical conductivity, BEC), and “hemoglobin” (calculated from BEC). Heritabilities were estimated using genomic relationship information obtained from 600k SNP chip data. All 13 parameters exhibited a significant change after 5 h of heat exposure and most did not return to pre-heat levels throughout the duration of the study. Eight parameters (base excess, glucose, hemoglobin, HCO3, hematocrit, K, pCO2, TCO2) had heritability estimates differing from zero at one or more time points (0.21 to 0.45). The traits with significant heritability would be good candidates for use as biomarkers in a selection program if they are correlated with traits of economic importance that are more difficult to measure. QTL were identified for nine of the traits at one or more time point. These nine traits, however, did not have significant heritability estimates suggesting that while some QTL have been identified their effects are generally small.
